Abstract

ABSTRACTAntagonistic Interaction between Lactobacillus 5NS and Candida tropicalis. Centrifuged supernatant of medium where Lactobacillus 5NS grown inhibited growth of three strains of Candida tropicalis. The Inhibition activity was detected using paper disc method. When the supernatant was induced with living cells of C. tropicalis Y393 strain, it inhibited the tested yeast strains. Inhibition activities of this supernatant against C. tropicalis Y390, Y391 and Y393 were equivalent to that of phenol at the concentration of 20.56 , 25.02 and 25.04 mg/ml, respectively. However when the supernatant was induced with heated or living cells of Candida tropicalis Y390 and Y391, it did not show any inhibition activity on all yeast strain tested. Thus the data indicates that there may be a specific interaction between Lactobacillus 5NS and C. tropicalis Y393 in which under the induction of specific certain yeast such as C. tropicalis Y393, Lactobacillus 5NS produces a toxin-like that is not spesific for the yeast target. Such interaction between normal flora and patogenic yeast may be happen in the digestive tractus.Key words: Lactobacillus, yeast, Candida tropicalis, antagonist

Abstract

ABSTRACTStudy of Some Bacteria to Heavy Metal Resistent (Hg, As, Cd, Ni, Pt and Se). The study was conducted to evaluate the resistance of some bacteria toward heavy metal (Hg, As, Cd, Ni, Pt and Se) by monitoring growth inhibition in Minimal Basal Medium (MBM) and Nutrient Broth (NB). Prior to growth inhibition study, pre selection of heavy metal resistance was determined by disk blanks methods. The results showed that AT 01, AT 03 and AT 11 were found to be heavy metal-resistent, however Pseudomonas sp. was a heavy metal-sensitive. The study also showed that AT 01 grew better in NB than MBM.Key words: Resistance, heavy metal, Pseudomonas sp.

Abstract

ABSTRACTThe Screening of Isolated Rhizobium from Wamena on Weki in Green House Condition. The green house experiment has been carried out to study the effectiveness of some Rhizobium isolates on the growth of weki (Paraserianthes falcataria) seedlings. Thirteen isolates of Rhizobium viz. 1wy(1), 1wy(2), 1wy(3), 2wy(4), 3wy(1), 3wy(2), 4wy(1), 5wy(1), 6wy(1), 7wy(1), 9wy(1), 10wy(3) and 11wy(1) have been screened and inoculated to the plant seedlings. The purpose of the study was to find out whether the Rhizobium isolates able to stimulate and improve the growth of weki seedlings. The results showed that all of the Rhizobium isolates could stimulate the production of the root nodules . However each isolate was difference effect on the plant growth. Isolate number 11 wy(1) could stimulate all the plant growth parameters significantly.Key words: Screening, seedling, Paraserianthes falcataria, weki, green house, Rhizobium.

Abstract

ABSTRACTEffect of Arbuscular Mycorrhizae Fungi and Phosphate Fertilizer to the Growth of Cashew on Red Yellow Podzolic Soil. The research about the effect of arbuscular mycorrhizae (AM) fungi and phosphate fertilizer applications to the growth of cashew seedling (Anacardium occidentale L.) grown on the red yellow podzolic soil was conducted in the green house at Cimanggu Research Installation, Research Institute for Spice and Medicinal Crops, Bogor. Completelly randomized design with factorial pattern, two factors and three replications was used. First factor was AM inoculation consisted of: without AM, Glomus etunicatum, Mycofer, whether second factor was phosphate fertilizer level consisted of: 0, 15, 30, and 45 g P2O5 per plant. The result showed an interaction between AM and phosphate fertilizer levels to the plant height, leaves dry weight, and spore population, and the best result on giving mycofer with P level 30 g P2O5/plant. AM inoculation significantly effected plant height, stem diameter, dry weight of leaves, stem and root, percentage of AM infection, and spore population, whether phosphate application significantly effected all of the parameters.Key words : Arbuscular mycorrhizae, phosphate, Anacardium occidentale L., plant growth

Abstract

ABSTRACTGrowth and Yield of Temoe Lawak in Polybag which Planting Material from In Vitro Culture. The growth and yield of temoe lawak (Curcuma xanthorrhiza Roxb.) from in vitro culture was conducted in the green house, Bogor from October 2000 to September 2001. The materials were obtained from temoe lawak plant two months after acclimatizating. The plant were planted in the polybag 60 cm x 60 cm. The treatments were consisted of the mixtures of soil and cattle manure with the proportional of 1 : 1 ; the soil and rice husk (1 : 1) ; the soil, cattle manure and rice husk (1 : 1 : 1). The experiment was designed according to randomized block design with three replicates and ten polybags per replicate .The parameters observed were plant performance, growth percentage at 5 and 10 months, rhizome weight per plant, length and width of primery rhizome, roots and water rhizome weight per plant. The result of experiment showed that the growth percentage of plant were 100% on ten months for all treatments. The growth and rhizome performance were similar with the parent. The best treatment for growth was the mixture of soil and cattle manure on the proportional of 1 : 1, with tiller numbers (3,57) , leaves number (18,2), plant height (140,6 cm) in five moths, rhizomes weight per plant (472,8 g), length and width of primery rhizome each 8,1 cm and 6,3 cm in ten months.Key words : Curcuma xanthorrhiza Roxb, growth, yield, in vitro

Abstract

ABSTRACTThe Stimulating Capacities of the Phosphate Solubilizing and the Nitrogen Fixing Microbes on the Maize Plant. The green house experiment has been carried out to study the effectiveness of the phosphate solubilizing microbes (PSM) and nitrogen fixing bacteria (NFB) on the growth of maize (Zea mays). The phosphate solubilizing microbes were Aspergillus sp. and Gigaspora margarita while the nitrogen fixing bacteria were Azospirillum sp. and Azotobacter sp. have been used in this experiment. The aim of the experiment was to find the suitable and effective inoculant on the maize and its expected the growth of the plant. The experiment used The Completely Randomize Design with three replicates. The treatments were uninoculated plant (control), the plants inoculated with single microbe of PSM, NFB and the combination of both. The results indicated that the phosphate solubilizing microbes (PSM) and the nitrogen fixing bacteria (NFB) which were either used in single or combination formula could stimulate the plant growth. The dual inoculation of Azotobacter sp. and Aspergillus sp. were found the higest yield of maize.Key words : The phosphate solubilizing microbes, the nitrogen fixing bacteria, Mycorrhiza vesicular arbuscular, Zea mays.

Abstract

ABSTRACTFeed Consumption and Digestibility Capacity on Cuscuses (Spilocuscus maculatus and Phalanger spp.). The research was conducted to know feed consumption and digestibility capacity on cuscuses in the Small Mammal Captivity on Zoology Division, Research and Developmment Center for Biology, Indonesian Institute of Sciences (LIPI), Bogor. The research material used were five cuscuses, each of them placed in individual cage. Species of cuscuses are Spilocuscus maculatus (two heads), Phalanger orientalis (one), P. gymnotis (one), and P. ornatus (one). The feedstuff is given in fresh form, consists of fruit, leaves, and extra feed. The fruit consists of passion fruit (Passiflora edulis), banana (Musa paradisiaca), guava (Psidium guajava), coconut (Cocos nucifera), and sweet corn (Zea mays). Leaves were kemang (Mangifera caesia), pohpohan (Pilea trinervia), and kangkung (Ipomea aquatica), and sweet potatoes (Ipomea batatas); while extra feed consists of bread and dog food. The result showed that the average of dry matter intake on cuscuses was 37.83 ? 6.94 g DM/kg BW0.75/head/day, while those on S. maculatus was 38.04 ? 6.25 g DM/kg BW0.75/head/day and on Phalanger spp. was 37.68 ? 7.57 g DM/kg BW0.75/day. The digestible coefficient of nutrient on cuscuses was 86.72% dry matter (DM), 66.86% ash, 69.86% crude protein, 95.51% ether extract, 73.58% crude fiber, and 92.09% nitrogen free extract. Total Digestible Nutrient (TDN) on cuscuses was 93.96 ? 3.22% and Digestible energy (DE) was 3.71 ? 0.39 kkal/g DM.Key words : Consumption, digestibility, Spilocuscus maculates, Phalanger spp., captivity

Abstract

ABSTRACTGenetic Diversity of Slow loris (Nycticebus coucang) based on Mitochondrial 12S rRNA gene. The research on genetic diversity of slow loris Nycticebus coucang (kukang) was carried out. The samples are 12 individuals from three locations (Sumedang and Jember in Java, and Lampung in Sumatera). Total DNA was extracted from blood and tissue. The mitochondrial 12S rRNA nucleotide sequences were determined to investigate genetic diversity of this species. This region was amplified by using L1091 and H 1478 primers by PCR. As a result of the analysis for 386 bp nucleotide sequence, five haplotypes were found, two from Java and three from Sumatra, respectively.Key words : Slow lories, Nycticebus coucang, genetic diversity, 12S rRNA mtDNA

Abstract

ABSTRACTThe Identification of Yeasts Which Were Isolated from Soils in the Wamena Biological Garden, Jayawijaya Residence, Papua Province. The present study deals with the isolation and identification of yeasts isolated from soil samples in The Wamena Biological Garden, Papua. Microbial population in 11 samples were estimated by plate count method. The yeasts population were ranged from 30.0 x102 until 16.5 x106 cells/g soil. Fifteen isolates of yeasts were taxonomically studied. On the basis of morphological and physiological characteristics these isolates were belonging to two group namely ascomycetous-imperfect yeasts and basidiomycetous yeasts. Futher, they were separated into three group. Out of 15 isolates, eleven isolates were included in group I and identified into genus level as Cryptococcus, three in group II as Candida and one isolate in group III identified into species level as Cryptococcus albidus. Type of sample sources appeared has no effect on yeasts diversity as shown by similar yeast isolate was observed isolated from different sources.Key words : Ascomycetous-imperfect yeasts, basidiomycetous yeasts, The Wamena Biological Garden, Papua.
